Prothiaden Tablet - Precautions & How to Use

Before using Prothiaden Tablet, inform your doctor about your current list of medications, over the counter products (e.g. vitamins, herbal supplements, etc.), allergies, pre-existing diseases, and current health conditions (e.g. pregnancy, upcoming surgery, etc.). Some health conditions may make you more susceptible to the side-effects of the drug. Take as directed by your doctor or follow the direction printed on the product insert. Prothiaden Tablet - Contraindications

Missing a dose

Overdosage of Prothiaden Tablet

  • Do not use more than prescribed dose. Taking more medication will not improve your symptoms; rather they may cause poisoning or serious side-effects. Questions - Prothiaden Tablet

Yes, Prothiaden may cause weight gain. Dosulepin is the generic name of Prothiaden. This medication is not FDA-approved due to low therapeutic index and significant toxicity due to overdose. Prothiaden inhibits the reuptake of biogenic amines, increases the available neurotransmitter levels at the synaptic cleft. Its use is only recommended for patients who are intolerant to alternative antidepressant therapies.

The earliest known antidepressants are Tricyclic antidepressants (TCAs), also called cyclic antidepressants. Though being effective, TCAs have been replaced by other antidepressants causing fewer side effects. However, in some cases, TCAs were seen to relieve depression when other treatments have failed.

A study published in PubChem found that patients taking Prothiaden and other antidepressants had a mean weight gain of 1.3-2.9 pounds per month depending on drug, dose, and duration over the course of 6 months. However, the exact weight gain varied, depending on the TCA used.

Some other reports indicate that TCA users experience more than 10 pounds (~4.5Kgs) of weight gain which may be due to increased cravings for sweets among people taking antidepressants.

However, one other study at Massachusetts General Hospital's psychiatry department demonstrated significant variations between different individual antidepressant strategies in their propensity to contribute to weight gain. According to researchers, the magnitude of weight gain is modest, so the clinicians should prefer treatments according to specific patient preference or in people for whom weight gain is a particular concern.

TCAs are not the only class of antidepressants that may have side effects of weight gain. Other antidepressants and piperazinyl phenothiazines such as SSRIs (Brisdelle/Luvox) and MAO inhibitors (Nardil/Parnate), may also cause weight gain.
